If you killed her before the Help Kyass quest then you will miss this long two/three parts quest, but you can resume it simply going to Kyass after speaking to Castle's Captain. And the Drath's Tome quest you can do as well without her assigning it to you, because you can find other characters to get the quest from and give the Tome to (Patrick castle). A certain level of reputation allows for additional dialog/quests. That is the only game-based concern, and it's not really a concern.
